In order to demonstrate the efficacy of POZ irinotecan, a number of mouse xenograft studies were done with HT-29, NCI-H460, A2780, IGROV-1, KB, and MDA-MB231 human tumor cell lines [60]. In one study, the HT-29 colorectal tumor fragments were implanted subcutaneously in the flank of female athymic Ncr:Nu mice and all the tumors were allowed to grow to an average weight of 150 mg. Each group of mice (n=10) was intravenously dosed every fourth day for a total of 3 doses of irinotecan or POZ-irinotecan at 40, 60, or 90 mg/kg. The control group received normal saline. The animals were weighed and the tumors measured twice weekly after administration of the first drug injection. All doses of irinotecan and POZ-irinotecan were well tolerated. The maximum loss in body weight for irinotecan and POZ irinotecan was 8% and 2%, respectively. The effects of the test compounds on HT-29 tumor growth are shown in Fig. 6.13.
